Evidence for a role of tyrosyl residues in cell membrane permeability.

چکیده
In the presence of tyrosinase, red blood cells are Ipsed. Lysis follows an enzyme-mediated reaction, with the amount of lysis being proportional to the tyrosinase concentration. Cyanide, which inhibits tyrosinase, prevents lysis of the red blood cells by tyrosinase. Heat-denatured tyrosinase has no effect on red blood cells. Tyrosinase also exhibits a pronounced effect on the permeability of isolated rat diaphragms. Glucose uptake is inhibited 28 to 44 % whereas glycine uptake is stimulated 50% by tyrosinase. Ehrlich ascites cells, in the presence of tyrosinase, show decreased viability as measured by eosin dye uptake. These data strongly suggest that the tyrosyl residues of the protein layers of the cell membrane are involved in the maintenance of cell membrane integrity.

Oxidative membrane damage and its involvement in gamma radiation-induced apoptotic cell death.
Background: Recent results have provided increasing evidence to support involvement of membrane damage in the mechanism of ionizing radiation induced killing of mammalian cells. These findings have stimulated renewed interest in evaluating the damage to membrane as a primary initiator in radiation-induced cell killing especially in apoptotic death. The present study was aimed to gain deeper ins...
متن کاملEvidence for Histidine Residues on Plasma Membrane Phosphatidate Phosphohydrolase from Rat Liver
Objective(s) Phosphatidate phosphohydrolase (PAP) catalyzes the dephosphorylation of phosphatidic acid to yield Pi and diacylglycerol. Two different forms of PAP in rat hepatocyte have been reported. PAP1 is located in cytosolic and microsomal fractions and participates in the synthesis of triacylglycerols, phosphatidylcholine, and phosphatidylethanolamine, whereas the other form of phosphati...
متن کاملAn evidence for a potassium channel in endoplasmic reticulum based on single channel recording in bilayer lipid membrane
Introduction Numerous studies have demonstrated the presence of potassium selective channels in membranes internal organelles. These channels are essential to a large variety of cellular processes including intracellular 2+ a signaling, protein recycling, charge neutralization and cell protection. In contrast to the sarcoplasmic reticulum + here potassium channels have been clearly ...
متن کاملThe Role of Nanoparticle in Brain Permeability: An in-vitro BBB Model
Membrane permeability and P-glycoprotein (P-gp) efflux system are regulating factors in the drug brain penetration. Recently, some drug delivery systems have been developed to overcome these limitations. In this study, Metoclopramid has been encapsulated in PLGA nanoparticles using the emulsification/solvent evaporation technique for in vitro evaluation of the effect of PLGA nanoparticles on BB...
